چکیده

Grape ripe rot (Colletotrichum viniferum) causes huge losses in grape production vineyards southern China. However, the molecular mechanism against species and responsive genes implicated these processes are relatively unknown. Here, we present transcriptome analysis of berries from a C. viniferum-resistant (Vitis davidii Föex). Uninfected at day zero were used as control samples (CK), an inoculation was made zero, subsequently analyzed 1 day, 3 days, 7 days post (dpi), which exhibited sequential disease-progression stage. There total 1810 differentially expressed genes, including 1315 up-regulated 495 down-regulated transcripts. At dpi, (DEGs) predominantly enriched berries. In addition, viniferum-infected fruits considerable changes gene expression induced, those involved MAPK cascade, calcium ion binding, serine/threonine kinase activity enriched. According to our KEGG pathway analysis, numerous biological processes, such plant–pathogen interaction, phenylpropanoid biosynthesis, metabolism, grape–fungus interactions. Our research also revealed alterations pattern phenylalanine-pathway-related transcription factors (TFs) genes. We proposed model viniferum invasion produces intracellular extracellular Ca2+ deregulation stimulate activate TFs’ (WRKY, ERF, MYB) up-regulation, thus initiating disease-resistant responses tolerant Vitis species. results offer comprehensive transcriptomic data about grape, will aid understanding underlying plant viniferum.

Transcriptome analysis of genes involved in anthocyanins biosynthesis and transport in berries of black and white spine grapes (Vitis davidii)

BACKGROUND The color of berry skin is an important economic trait for grape and is essentially determined by the components and content of anthocyanins. The fruit color of Chinese wild grapes is generally black, and the profile of anthocyanins in Chinese wild grapes is significantly different from that of Vitis vinifera. However, V. davidii is the only species that possesses white berry varieti...

Transcriptome Analysis of Nicotiana tabacum Infected by Cucumber mosaic virus during Systemic Symptom Development

Virus infection of plants may induce a variety of disease symptoms. However, little is known about the molecular mechanism of systemic symptom development in infected plants. Here we performed the first next-generation sequencing study to identify gene expression changes associated with disease development in tobacco plants (Nicotiana tabacum cv. Xanthi nc) induced by infection with the M strai...

Identification and localization of the bilitranslocase homologue in white grape berries (Vitis vinifera L.) during ripening

A homologue of the mammalian bilirubin transporter bilitranslocase (BTL) (TCDB 2.A.65.1.1), able to perform an apparent secondary active transport of flavonoids, has previously been found in carnation petals and red grape berries. In the present work, a BTL homologue was also shown in white berries from Vitis vinifera L. cv. Tocai/Friulano, using anti-sequence antibodies specific for rat liver ...

An immunohistochemical study of the compartmentation of metabolism during the development of grape (Vitis vinifera L.) berries.

The compartmentation of key processes in sugar, organic acid and amino acid metabolism was studied during the development of the flesh and seeds of grape (Vitis vinifera L.) berries. Antibodies specific for enzymes involved in sugar (cell wall and vacuolar invertases, pyrophosphate: fructose 6-phosphate phosphotransferase, aldolase, NADP-glyceraldehyde-P dehydrogenase, cytosolic fructose 1,6-bi...

Identification and Characterization of New Record of Grape Ripe Rot Disease Caused by Colletotrichum viniferum in Korea

In 2016, grape fruits showing ripe rot symptom were found in fields of Korea. The fungus was isolated and identified as Colletotrichum viniferum based on morphological characteristics and nucleotide sequence data of the internal transcribed spacer, glyceraldehyde-3-phosphate dehydrogenase and β-tubulin. To our knowledge, this is the first report of C. viniferum causing grape ripe rot disease of...
